Land Rover 109 4.6 engine knock

My brother has a brand new 4.6 engine fitted to his 109.He has a lightish knock when the engine is warm.He fitted a cam retention plate which made no difference.He reckons one spark plug is cleaner than the rest.Could this be a slipped liner causing the knock?It is fitted with the twin SUs it had originally[rejetted for the 4.6.
