I have my fully rebuilt 3.5 rover engine up for sale at the moment. I want to replace it with a 4.6 rover. Is there anything I need to be aware of? I'm presuming everything on the 3.5 will fit on the 4.6? Any advice greatly appreciated.

The 4.6 comes in serpentine/intermediate and pre-serpentine variants, only the pre-serpentine will fit your existing front cover. You'll need to sort out the clutch and everything downstream, and of course the fuelling requirements will be different.

It's a fairly straight forward swap. If you are using the timing cover, oil pump, dissy off the 3.5 you will need to fit a spacer to take up the extra crank length. These are available from most RV8 specialists. The 3.5 oil pump needs to be in good condition.
