Given that you presumably already have: A toothed wheel on the crank An EDIS module 2 coil packs - Mounted and correcct length leads I'd have though it would be much easier (possibly cheaper) to simply add in an ECU to control the ignition. When installed in the vehicle how does it behave if you take a live feed directly from the battery? If that works OK it would point to a problem in the wiring anywhere between the battery to the coil via things like the ignition switch, fuse box and any connectors. Is the piston actually moving up/down the bore? If I understood correctly you have the head of so presumably no big hole in the crown? If the valves are not obviously open you should be getting some compression even at cranking speed, hard to imagine losing all your air via a partially open valve, ...

Photo would be a great idea. Personally I'd go down the electronic route, much more flexible and should be possible to easily re-calibrate. My car has a mechanical speedo but since building I've changed the 'box which has different speedo' gearing and the diff' ratio so now reads out, I'm currenlty ...

I only have experience of the 2WD version of the box and I think they differ in that area but I'd say you'd struggle to mix and match. On the 2WD the rear case is significantly different, a fair bit larger. I think this is due to the reverse gears being sited in that casing on the R380 IIRC this is...

Just had a look at the RPi offering. Looks like they sell it as a conversion (at £4.6k) which seems to be specific to the MGB.
